Bio-ethanol can be a product from an edible source which known as first generation bio-ethanol such as sugarcane and corns. Second generation of bio-ethanol is from lignocelluloses biomass is from non-edible source such as palm oil biomass. This make second generation of bio-ethanol is a better choice to replace fossil fuel without disturbing food sources. By converting the waste into valuable product we can reduce the environmental problem. Lignocelluloses material contain about 50% 60% carbohydrate in the form of cellulose and hemicelluloses which may be fermented to ethanol and 20%-35% of lignin (Mats Galbe et al, 2007). Nowadays palm oil tree is one of the main source for the production of bio-ethanol but there are not m uch attention is been given to oil palm sap which are product that come from the tree component such as oil palm trunk and oil palm fronds. Malaysia is the world’s largest exporter of palm oil product. Malaysia produces a large amount of agro-industrial residue with oil palm industry itself produced about 33 million tons of residues annually in the form of empty fruit bunch, fiber and shell (MPOB, 2009; Mohamed and Lee, 2006). Malaysia has produced about 51 million tons of oil palm fronds with 53% of the total palm biomass in year 2008 (Goh et al, 2010; MPOB, 2009). Bio-fuel that produces from palm oil tree is an environmental friendly therefore many interest are being shown to this sources. Mostly, the major parts of the solid biomass from the oil palm tree are being leave behind on the plantation is to be found as oil palm fronds. It has been reported that about 46837K tons of oil palm fronds has been produced in Malaysia in the year 2007 as an agriculture wastes. Palm oil frond is one of the useful raw materials for the production of bio-ethanol which is environmental friendly way. Pre-treatment of the oil palm frond need to be done to achieve a good bio-ethanol production. However, ethanol production from lignocellulosic biomass is relatively expensive because of the latest technologies. The main contradict are low yield and cost of the hydrolysis process (Sun and Cheng, 2002). Thus, oil palm frond juice is being introduced as another source for the production of bio-ethanol. Previous study has been shown that oil palm juice is suitable to used as fermentation feedstock because there was no inhibition on microbial growth or product formation, there were no impurities, it was easy to be operated, and it had no risk on health and safety (Zahari et al, 2012). Oil palm frond sap has been proved by Zahari et al. (2012) contains high sugar content which is 76.09 Â ± 2.85 g/l. There has been reported by Eze and Ogan (1988) that sucrose is the dominant sugar in the oil palm frond sap that consist of 10% w/v, as for glucose and fructose consist of et al, 2010. However in Malaysia, a study shows that glucose is the dominant sugar in the oil palm sap (Kosugi et al., 2010). Since there are many studies show that oil palm juice contains higher sugar content, a high level of the production of bio-ethanol from oil palm sap is further study. A study by Nwachukwu (2008) shows that improving yeast resistance by protoplast fusion increased yields of ethanol by 16% v/v. Apart from that, types of fermentation also one of the factor that improve the production of bio-ethanol. Thus, this project aim is to increase the yield of the bio-ethanol production using oil palm frond juice by various type of fermentation process. PROBLEM STATEMENT Uses of natural sources like petroleum is been used over the decades and the demand of this sources are being increases from time to time. Over the century, there are an increasing of energy consumption with the increasing of world population, thus more growing countries become industrialized which there are possibility that the sources will be depleted one day. Apart from that, petroleum is harmful to human and environment and with the increasing of fossil fuel will lead to increasing of carbon dioxide that eventually leads to global warming issue. Thus, an alternative sources of the fuels is been quest to overcome the shortness of the fossil fuels. Bio-ethanol is one of the solutions to this problem. This is because bio-ethanol is a form of renewable energy source which are easy available, low cost and most efficient bio-fuel. Chooklin et al, 2011) Oil palm frond (OPF) will be harvest from oil palm tree from a local oil palm plantation. The leaves from the frond will be cut off and discard. The harvest OPF will be press using sugar-cane machine press as soon as possible after the frond was harvested. OPF juice produced will be collect and filtered using coffee filter. The filtered OPF juice will be centrifuged at 10,000 rpm for 15 minutes. Supernatant will be stored at -20Â °C before use. 4.2 OPF Juice Sugar Composition (Ethanol Production Using Immobilized Saccharomyces cerevisiae in Lyophilized Cellulose Gel by Eleonora Winkelhausen et al, May 2010) HPLC type:HPLC (Shimadzu Japan ), equipped with refractive index Detector Type of column: APS-Hypersil column ( diameter of 250mm x 46mm) Mobile phase:70% Acetonitrile and 30% de-ionized water Column temperature:40Â °C with maximum operating temperature of 80Â °C Flow rate: 0.6 ml/min Sample volume: 20Â µl Pressure: 10MPa 4.3Â  Pure Culture Establishment (Study on Bioethanol Production from Oil Palm Trunk (Opt) Sap by Using Saccharomyces Cerevisiae Kyokai No.7 by Nina Farhana 2010) (Isolation of Microorganism from Oil Palm Sap by Nurul Nadia Ummira, 2011) 100mL of the Nutrient Broth Medium supplemented with g/L; Yeast Extract, 5; Peptone, 10 and 15% of glucose solution, 50 (Jamaludin, 2010) will be measured in a 250mL conical flask and will be autoclaved at 121 C in 15 minutes. Saccharomyces cerevisiae will be subculture into the Nutrient Broth. The mixture will be incubated for 24 hours at 150 rpm, 30oC. The suspension obtained will be subculture on the Nutrient Agar slant and will be incubated for another 2-3 days. The Nutrient Agar slants will be stored in refrigerator at 4oC. 4.4 Inoculums preparation (Study On Bio ethanol Production From Oil Palm Trunk (OPT) Sap by Using Saccharomyces Cerevisiae Kyokai No.7 by Nina Farhana 2010) (Optimization study of ethanol fermentation from oil palm trunk, rubber wood and mixed hardwood hydrolysates using Saccharomyces cerevisiae by K.L. Chin et al , 2010) About 3 4 loop of pure culture will be transferred from agar slant into 100 mL of sterile Nutrient Broth in 250 ml conical flask plug with sterile cotton. The flask will be incubated for 24 hours at 30Â °C and 150 rpm until reach standard initial concentration. The cell concentration will be standardized to 0.2 0.4 g/L (OD = 4.2) determined by turbidimetry at 600 nm. 4.5 Fermentation a) Batch fermentation will be performed for the optimization of bio-ethanol production. b) 20% (v/v) of inoculums size will be inoculated into the bioreactor with the condition of pH 4.5 and temperature of 30Â ºC , air flow rate of 1 vvm, agitation of 200 rpm with the initial 02 concentration of 30 %. c) The fermentation process will be carried out for 42 hour and sample will be taken out every 3 hour and analyze for bio-ethanol production, sugar and cell biomass. d) All experiment will be run in triplicate. 4.5.1Â  Batch fermentation. 4.5.1.1Â  Effect of sugar concentration (Effect of Cultural Conditions on Ethanol Production by Locally Isolated Saccharomyces Cerevisiae Bio-07 by Arifa Tahir et al, 2010) OPF juice volume use will be varied in the range of 30% to 50%. (30%, 40%, 50%) 4.5.1.2Â  Effect of agitation speed (Optimization of Fermentation Medium for the Production of Ethanol from Jaggery Using Box-Behnken Design by Mary Anupama.P et al 2010) The fermentation will be carried out at different agitation rate in the range of 125 rpm to 175 rpm. (125, 150, 175) 4.5.2Â  Fed Batch fermentation 4.5.2.1Â  Effect of feeding rate (Production of Ethanol by Fed- Batch Fermentation by Ngoh Gek Cheng et al, 2009) The fed batch fermentation will be run according to the best optimized condition from the batch fermentation condition. Substrate will be feed continuously into the bioreactor using peristaltic pump at glucose concentration (glucose concentration from OPFJ?) varying from 2 ml to 8 ml at feeding rate of 2 ml/h . (2 ml , 4 ml, 8 ml) 4.5.2.2Â  Effect of feed time (Production of Ethanol by Fed- Batch Fermentation by Ngoh Gek Cheng et al, 2009) a) Substrate will be feed from the interval of every one hour to three hour at feeding rate of 2 ml/h. ( 1 hour , 2 hour , 3 hour ) 4.6Â  Harvesting (Study on Bioethanol Production from Oil Palm Trunk (Opt) Sap by Using Saccharomyces Cerevisiae Kyokai No.7 by Nina Farhana, 2010) Each batch of fermentation will be terminated according to optimized time period. b) Sample (15 ml) will be taken out and the samples will be kept in refrigerator at 4oC before analyze for bio-ethanol production, sugar and cell biomass. 4.7Â  Yeast strain and it improvement (High-level Production of Ethanol during Fed Batch Ethanol Fermentation with a Controlled Aeration Rate and Non-Sterile Glucose Powder Feeding of Saccharomyces cerevisiae by Hyen-beom-seo et al, 2009) a) Saccharomyces cerevisiae will be growth on Nutrient Broth (yeast extracts, 10g/l; peptone, 20g/l; glucose, 20g/l and agar, 20g/l) slant in a 30 ml universal bottle. b) The slant culture will be exposed with UV light (6 watt, 254nm) for 15 second interval at a distance of approximately 7 cm from the slant. c) A loopful of irradiated slant culture will be streak onto a Nutrient Broth agar plate and subsequently incubated for 3 days at 27Â ºC. d) After that the colonies will be incubated in Nutrient Broth medium containing 100 g/l of ethanol at 27Â ºC while shaking at 100 rpm for 5 days to select resistance colonies. 4.8 Analysis method 4.8.1 Sugar content by HPLC (Oil Palm Fronds Juice as Future Fermentation substrate: A Feasibility Study by Che Mohd Hakiman Che Maail et al, 2014) HPLC type:HPLC (Shimadzu Japan ), equipped with refractive index Detector Type of column: APS-Hypersil column ( diameter of 250mm x 46mm) Mobile phase:70% Acetonitrile and 30% de-ionized water Column temperature:40Â °C with maximum operating temperature of 80Â °C Flow rate: 0.6 ml/min Sample volume: 20Â µl Pressure: 10MPa 4.8.2Â  Cell dry determination (Study on Bioethanol Production from Oil Palm Trunk (Opt) Sap by Using Saccharomyces Cerevisiae Kyokai No.7 by Nina Farhana 2010) Cell suspension will be vaccum-filtered using 0.45Â µm filter paper Cell suspension for each samples will have to be rinsed 2x with distilled water. The possibilities of extra job creations in recycling industries are taken away by the costly incineration technologies. In a way, it is a colossal waste of not only natural resources but public money. (Recycling versusà ¢Ã¢â€š ¬Ã‚ ¦) It is a myth that incinerators produce energy from waste but the truth is that by burning the waste they increase the consumption of fresh materials. This tends to increase the energy consumption in producing those materials. Energy consumption is at least 4-5 times more in producing fresh material in compare to recycling of those materials. (Burning wasteà ¢Ã¢â€š ¬Ã‚ ¦) Residual ash generated is almost found to be 30 percent of the original waste by weight and that needs to be disposed of to landfill sites. Several towns such as Canberra in Australia by 59%, the city of Edmonton in Canada by 70% and the towns in New Zealand and USA have successfully achieved reductions in land filling up to even 70 percent without resorting to any incineration technology. This has been achieved by them in a short span of 5 years. As such, most of the people in the industry have the view that 60% is quite realistic target for reduction in landfill by recycling and other methods. Then him and his family moved to Leipzig from the anti-Semitic riots. His ambition in his childhood had been a career in law but then he decided to be medical student before he entered to Vienna University in 1873. After this he desire to study natural science and to solve challenging problems that confronted contemporary scientist. In his three year at Vienna University Freud began his research in central nervous system in the physiological lab under the direction of German Physician Ernst Wilhelm Von Brucke. In 1881 after completing a year compulsory military service he receive his medical degree. After he received his degree he remained at the university as a demonstrator in the physiological laboratory. Freud spent three years at the General Hospital of Vienna devoting himself to psychiatry, dermatology, and nervous disease. In 1885 after appointed as lecturer in neuropathology at the university he decided to leave his post in the hospital. Later that same year Freud studies under Jean Charcot in which centered largely on hysteria, influenced Freud greatly in channeling his interest to psychopathology. Summary of The Spire

Jocelin, Dean of a medieval cathedral, has had a vision which he believes reveals that he must add a four hundred foot spire to the cathedral. The decision is a controversial one, especially as the work proves disruptive and the master builder, Roger Mason, discovers that the building lacks the requisite foundations to support the spire. Jocelin is insistent that faith will be sufficient and accuses the master builder of being timid, and of playing for time in order to keep himself and his men in employment. Jocelin is maintained in his belief that the spire will stand by the news that his bishop is sending a Holy Nail (from the crucifixion) from Rome to protect the spire. The cathedral's caretaker, Pangall, hates the disruption and the workmen's mockery which he suffers. There are early hints that he is impotent. Jocelin is horrified when he notices that Roger and Goody, Pangall's wife, are sexually attracted to each other. However, he realises that, if he does not intervene, their adultery will prevent Roger from leaving. Roger's wife Rachel reveals that she and her husband are childless because she finds sex makes her laugh. Jocelin climbs to the roof to inspect the work and finds it exhilarating. However, he has what is eventually revealed to be tuberculosis of the spine, and this illness gradually becomes worse. He is also increasingly troubled by sexual dreams relating to his own attraction to Goody. A pit has been dug in order for the master builder to look for foundations, and there is a crisis when the earth in it is seen to be creeping. The stones start to make a high-pitched whine and to splinter. Roger wants to stop work, but Jocelin forces him to continue. The result is that the anxious workmen become a mob and – as only later becomes apparent – they pursue and murder Pangall, burying his body beneath the cathedral pavement as a pagan charm to keep the spire from falling. Jocelin becomes increasingly obsessed with the spire, shutting out all other concerns. However, he feels guilty about Goody and tries to speak to her. When she sobbingly rejects his approaches, he climbs the spire to seek solace. It is revealed that Goody is pregnant. Roger becomes increasingly sullen and unpopular and the singing of the stones becomes worse, so that the master builder again begs Jocelin to halt the work. He paints a persuasive picture of the spire's collapse, but Jocelin resists and makes him continue. Shortly after this, Jocelin climbs the spire and witnesses Roger and Goody having sex. A steel brace is made and fitted to the spire. While this is going on, Jocelin secures a place for Goody in a local convent. But when Rachel discovers Roger's infidelity, she attacks first Goody, then Roger, and Goody dies in childbirth. Jocelin becomes more unwell and is tormented by remorse and sexual feelings, although he is relatively happy when helping the workmen. Roger becomes an alcoholic and has a breakdown. At midsummer, Jocelin realises that the workmen have left their work to attend pagan festivities. More Summary of Devil at My Heels The spire nears completion as the Holy Nail approaches. Jehan, Roger's second-in-command, now in charge of the work, miscalculates and damages the spire. An official from Rome, referred to only as the Visitor, interviews Jocelin and relieves him of his authority. In a raging storm, Jocelin climbs the spire and hammers the Nail in place, after which he has two mystical visions of Goody. Jocelin's aunt, Lady Alison, visits him and reveals that he was only appointed Dean thanks to her. The dumb sculptor, Gilbert, shows Jocelin that the pillars supporting the cathedral roof are not solid but filled with rubble. Father Adam reads aloud Jocelin's sermon describing his original vision of the spire. The process of Jocelin's disillusionment continues when Anselm – Jocelin's former teacher but now his junior as the Sacrist of the cathedral – denies that they were ever really friends. Jocelin goes to seek Roger's forgiveness, on the way having two mystical revelations inspired by an appletree and a kingfisher. He is briefly reconciled to Roger before the master builder becomes angry and throws him out. On the street, Jocelin is set upon by a mob. Nearing death, Jocelin has his effigy sculpted, and finally has several intimations which seem to explain his past experiences, and perhaps the whole of life itself. His dying thought is of the appletree, but the priest attending him, Father Adam, chooses to believe that Jocelin was in his dying breath murmuring the name of God. On one level this is a novel about the building of a spire upon a cathedral, the foundations of which are nothing but marsh and brushwood. It is about the resilience of those foundations against all odds; they hold a spire some four hundred feet high when, by rights, this shouldn't be possible. It follows the lives of a range of people involved in the building project, from the anonymous army of labourers who do the actual work at one extreme to the man who believes that God has chosen him to bring this work to a conclusion at the other. The spire stands at the end of the novel but it has destroyed the lives of Dean Jocelin, whose vision was the inspiration for its building; of Roger Mason, the master builder; of Roger's wife, Rachel; of Pangall's wife, Goody and the child she bears to Roger Mason. The reader is never confident whether the spire is the work of God or the work of the devil; what is clear is it is built upon human misery, upon argument and dissension within the cathedral's community, and upon the deceit of Dean Jocelin who holds high office in the church. As the spire reaches upwards the Dean feels its weight upon his back until he ends bent double; though he isn't aware of it, Jocelin suffers a crippling spinal disease – tuberculosis of the spine – which eventually kills him. Throughout the building of the spire he believes that the early physical manifestations of his condition are the visitations of his own guardian angel. The novel is also concerned with sexuality. It is about two men's desire for a simple woman who remains unaware of her own attractions; one, the Dean, suppresses his desires and suffers because of it, the other, Roger Mason, fulfils his desires, gets her pregnant and also suffers by it. Goody Pangall dies in childbirth; her husband is bullied mercilessly by Roger's men and finally driven cruelly to his death; Roger tries to kill himself and ends his day in madness; Rachel is left desolate, tending to her husband as if he were the baby they never had; Jocelin dies in both physical and spiritual agony. Finally the spire is left, a symbol but a very ambiguous one. Of the power of God to work miracles? Of a bargain with the devil? Or of the skill and endurance of man? Golding doesn't tell us.

Lenin proves this because he was feared and therefore people obeyed him. Another example of the use of cruelty is the cruel acts of Joseph Stalin.Stalin ruled with an iron fist and struck fear into peoples hearts. For example: Karl Marx imposed his theory of communist-based Marxism on his state because he thought it was the best thing for the state. The Communist League explains â€Å"Marxist-Leninist accept the concept of social class put forward above, but hold that a person's social class is determined not by the amount of his wealth, but by the source of his income as determined by his relation to labor and to the means of production. (Marxism and Class, 1) Marx created this intricate system of classes in order to maintain order thin his state. Although it was not successful, Marx created this policy for the well being of the state, so that the state could flourish. This relates to Machiavellian theory because Marx did as Machiavelli advised. He had the well being of the state in mind throughout his time of ruling. Another ruler who followed Machiavellian theory on well being of the state was Vladimir Lenin. Lenin not only followed a Marxist trend in his state, but he also tried to improve his state's economy.BBC states, â€Å"He introduced the New Economic Policy, here a measure of private enterprise was again permitted, a policy that continued for several years after his death. † (BBC,I) Lenin introduced this policy in order to preserve the economy of his state. It lasted several years after his death, which means he was successful in doing so. Lenin laid out a structure for the state to thrive because he had the well being of the state in mind while making his decisions. This follows Machiavellian theory because Machiavelli said to make every decision based on the well being of the state.Although he w as cruel and used militaristic power, he was ultimately trying to better the state in his own way which is exactly what Machiavelli preaches. All in all, Karl Marx, Joseph Stalin, and Vladimir Lenin were embodiments of the theories Machiavelli introduced in â€Å"The Prince†. They ruled and based their states on militaristic power, they ruled their states through cruelty and pain over compassion and love, and kept the well being of the state in mind in every decision they made. Machiavellian theories Justified their actions in that they each very closely followed them.Karl Marx had a strong military and had the well being of the state in mind, throughout his time as ruler, and Vladimir Lenin had a strong military and also made sure to keep the well being of his state in mind. Although the states of these rulers were not very successful and resulted in the deaths of many people, they were supported by the Machiavellian theories. The lesson to be learned is that although their actions were supported by the Machiavellian theories, they caused severe damage and terror to the world and these theories should ultimately not be used in any type of modern government.
